Introduction

Gene detection, also known as molecular detection, refers to a series of techniques that are adopted to test chromosomes, DNA and RNA in blood, tissues, cells and relevant samples based on different technology strategies. With revolutionary technical development at present, new gene detection techniques marked by the next generation sequencing have become cutting-edge technology of biomedicine. Moreover, they play an extraordinary role in the fields of life sciences research and applications.

Featured with accuracy, sensitivity, rapidness, simplicity and convenience, gene detection is utilized to detect various diseases and infectious agents, and guide disease prevention and precision treatment. Without a doubt, it is the core technology that is indispensable in the field of modern precision medicine. Additionally, gene detection is mainly applied in detection of different infectious agents, personalized detection of tumors, blood screening, antenatal diagnosis and genome analysis on drug metabolism, etc.
Classification

According to principles of detection technology, extensively applied gene detection primarily consists of gene sequencing, PCR based on nucleic acid amplification, FISH based on fluorescence in situ hybridization, gene chip technology and mass spectrometry, etc.. Among them, PCR, nucleic acid hybridization and DNA sequencing are three major foundations of gene detection.
Nowadays, polymerase chain reaction (PCR) is the most extensively applied gene detection technology. It is a new molecular biological technique invented by Mullis in the 1980s. Compared with traditional detection methods in the fields of biochemistriy, bacteriology, virology and serology, etc., PCR is strongly specific, highly sensitive, easy to operate, rapid and efficient. In terms of pathogeny, it can be used to detect pathogene and strains of pathogeny in different types or the same type. Particularly, it is suitable for detection of viruses and bacteria that cannot be easily cultured.
PCR mainly consists of conventional PCR and real-time PCR. Moreover, real-time PCR has been widely applied in detection of various causes of diseases, disease diagnosis as well as tumor and genetic disease screening, etc. thanks to its advantages (e.g., being highly sensitive, quantitative, simple, convenient and safe).
Methods

For example, if real-time PCR (probe method) is utilized to test DNA samples, the corresponding detection method mainly consists of the following steps:
1、Primer and probe design: Highly conservative segments are selected from a genomic sequence to design primers and probes. The probe includes a fluorescence reporter gene and a quencher.
2、Template preparation: Nucleic acid can be extracted manually or by a self-reacting device.
3、Reaction system construction and optimization: Parameters such as components, buffer concentrations and pH values should be all optimized continuously.
4、Amplified reaction procedure: Denaturation temperature, annealing temperature, elongating temperature and corresponding time should be tested and optimized one by one.
5、Result analysis: Standard curves are portrayed to realize determination according to relative and absolute quantification methods.
Characteristics

Now, a process platform that detects products based on conventional PCR and real-time PCR has been established. In addition, a multiplex PCR detection technique is also successfully developed.
